This is done on the serum of patient.
Procedure
serial dilution of the patient serum is taken 1:40 to 1:320
Now add equal volume of Salmonella antigen
This can be done as Slide method or as Tube method.
When running in the tube then incubate tubes for 12 hours or overnight.
Result
The highest dilution of the serum is noted where there is agglutination.If it ends in 1:320 then that is the titer.
Single test of 1:100 or more of O and 1:200 or more of H antigen is significant level.
A rising titer is more significant than single test ,which is 4 fold in a period of 7 to 10 days.
This test will be positive after 7—10 days of Enteric fever.
In the acute stage Antigen O will be postive and titer will be more than 1:160.This antigen appear early and also disappear early.
Antigen H rises late and disappear late.This will be positive in the recovery stage.
Antigen Vi is indicator of carrier stage.
